Contenuti | Il corso si integra con i moduli di Laboratorio di Biologia Molecolare e di Microbiologia per un comune obiettivo simulante parte di un progetto di ricerca: "Isolamento e clonaggio di un gene umano". Il Laboratorio di Genetica si occuperà in particolare di illustrare ed effettuare l'isolamento del DNA da genoma umano, valutata la concentrazione e purezza mediante analisi spettorofotometrica ed elettroforesi su gel d'agarosio. Da questo DNA sarà isolato e clonato un gene in un vettore plasmidico (nel Laboratorio di Biologia Molecolare ) che sarà utilizzato per trasformare batteri competenti (nel corso di Laboratorio di Microbiologia). |

Programma del corso | Introduzione alla intera esercitazione. Introduzione all’uso delle pipette. Isolamento del DNAgenomico. Valutazione della concentrazione e della purezza del campione allo spettrofotometro (A260, 230 280) Elettroforesi su gel di agarosio del DNA estratto. |

Contents | The course integrates with the Laboratory of Molecular Biology and Microbiology modules for a common goal simulating part of a research project: "Isolation and cloning of a human gene". The Genetics Laboratory will focus on illustrating and carrying out the isolation of DNA from the human genome, assessing the concentration and purity by means of spectrophotometric analysis and agarose gel electrophoresis . A gene will be isolated and cloned from this DNA into a plasmid vector (in the Molecular Biology Laboratory) which will be used to transform competent bacteria (in the Microbiology Laboratory). |
